چکیده

Results. Significant neuronal death was detected after 30 min (but not 10) of anoxia-aglycaemia (40 (4) vs 14 (5)% of control, P<0.05). At 30 min, phosphorylated ppFAK content was significantly decreasedby anoxic glucose-free conditions (55 (27)%of control, P<0.05). This effectwasmarkedly attenuated by thiopental (10 and 100 mM) and isoflurane (1 and 2%). Under control conditions, thiopental (1, 10, and 100mM) and isoflurane (0.5, 1, and 2%) increased ppFAK phosphorylation in a concentration-related fashion. This effectwasblockedbychelerythrin andbisindolylmaleimide I and IX (10mM, three structurally distinct inhibitorsof protein kinase C, PKC) but not the N-methylD-aspartate (NMDA) receptor antagonist MK801 (10 mM).

Thiopental and isoflurane attenuate the decrease in hippocampal phosphorylated Focal Adhesion Kinase (pp125FAK) content induced by oxygen-glucose deprivation.

BACKGROUND Thiopental and isoflurane exhibit neuroprotective effects against cerebral ischaemia. Here, we hypothesized that oxygen-glucose deprivation decreases the ATP-dependent phosphorylation process of Focal Adhesion Kinase (pp125FAK, a functionally important non-receptor tyrosine kinase), and that this phenomenon is attenuated by thiopental and isoflurane. METHODS Rathippocampal slices w...

Characterization of the postconditioning effect of dexmedetomidine in mouse organotypic hippocampal slice cultures exposed to oxygen and glucose deprivation.

BACKGROUND There is an increasing interest in the use of dexmedetomidine for anesthesia and sedation. Here, we used the mouse organotypic hippocampal slice culture to investigate whether dexmedetomidine exhibits postconditioning properties against oxygen and glucose deprivation (OGD). The role of the focal adhesion and extracellular-regulated kinases pathways in these effects were examined in b...

The Effect of Eight- Week High-Intensity Interval Training on the Expression of Cardiac Angiostatin and Focal adhesion Kinase Proteins at Left Ventricle in Diabetic Rats

Background: Diabetes can increase cardiovascular disease by altering the equilibrium between angiogenic stimulants and inhibitors of angiogenesis. In this study, we investigated the effect of High-Intensity Interval Training (HIIT) on angiostatin and focal adhesion kinase (FAK-1) in left ventricle cardiomyocytes in diabetic rats. Methods: In this experimental study, 24 male Wistar rats were ra...

Effects of dexmedetomidine on hippocampal focal adhesion kinase tyrosine phosphorylation in physiologic and ischemic conditions.

BACKGROUND Dexmedetomidine is a potent and selective alpha2-adrenoceptor agonist that exhibits a broad pattern of actions, including sedation, analgesia, and neuroprotection. Some of these actions (e.g., neuroprotection) may require targets involved in long-term cellular changes. The authors hypothesized that dexmedetomidine increases the expression of active (autophosphorylated) focal adhesion...

Interplay of Phosphorylated Apoptosis Repressor with CARD, Casein Kinase-2 and Reactive Oxygen Species in Regulating Endothelin-1–Induced Cardiomyocyte Hypertrophy

Objective(s):  The role of the Apoptosis repressor with caspase recruitment domain (ARC) in apoptosis and in certain hypertrophic responses has been previously investigated, but its regulation of Endothelin-1 induced cardiac hypertrophy remains unknown. The present study discusses the inhibitory role of ARC against endothelin–induced hypertrophy. Results:In present study Endothelin treated car...
